CD32a/CD32b/CD32c Antibodies

Target Information

CD32 denotes Fc gamma receptor II family members that bind the Fc portion of IgG and modulate immune cell activation. CD32a is encoded by FCGR2A and is an activating receptor with an immunoreceptor tyrosine based activation motif that signals through Src family kinases and Syk, expressed on neutrophils, monocytes, macrophages, platelets, and dendritic cells. CD32b is encoded by FCGR2B and is an inhibitory receptor with an immunoreceptor tyrosine based inhibitory motif that recruits SHIP and SHP phosphatases, expressed on B cells, monocytes, macrophages, dendritic cells, and some basophils and mast cells, and it constrains antibody mediated activation to maintain tolerance. CD32c is encoded by FCGR2C and can function as an activating receptor depending on its open reading frame status, with expression largely restricted to natural killer cells and some monocytes, and it contributes to antibody dependent cellular cytotoxicity. Genetic polymorphisms in FCGR2A, FCGR2B, and FCGR2C influence IgG subclass binding, receptor signaling strength, and susceptibility to autoimmune disease, infection outcomes, and responses to therapeutic antibodies. Clinical interest includes using CD32 isoforms as biomarkers and as targets to tune effector functions in antibody therapies.
