Target Information

TIP1 encodes a membrane-associated S-acyl transferase that localizes to endomembrane compartments and functions broadly in plant cell expansion, with especially prominent roles in polarized tip growth of root hairs and pollen tubes; loss-of-function alleles (tip1-1 and tip1-2) cause short, frequently branched root hairs, impaired pollen tube growth, and overall growth defects, consistent with a requirement for TIP1 activity in both tip growth and diffuse (non-tip) cell growth. Structurally, TIP1 is an ankyrin repeat-containing DHHC cysteine-rich domain protein (a DHHC palmitoyltransferase family member), with multiple N-terminal ankyrin repeats that are thought to mediate protein-protein interactions and a conserved DHHC catalytic motif within a cysteine-rich region that binds palmitate and supports enzymatic S-acylation; in functional assays, TIP1 can substitute for a yeast S-acyl transferase mutant and chemical inhibition of protein acylation phenocopies the tip1 root-hair defect, linking its molecular activity to growth control. Biologically, TIP1-dependent S-acylation is expected to regulate the membrane association, trafficking, and/or spatial activity of proteins required for cell surface expansion and polarized secretion, providing a mechanistic basis for its impact on root hair morphogenesis and general plant growth, and is distinct from tonoplast aquaporin TIP1;1 and TIP1;2, whose complete loss produces only mild physiological effects under standard conditions.
