TIGIT expression was assessed within CD19 negative (CD19-) and CD19 positive (CD19+) subsets of human PBMCs by staining the cells with CD19 eFluor™ 450 (Product # 48-0199-42) and 0.25 µg of TIGIT Recombinant Mouse Monoclonal Antibody (MBSA43), Alexa Fluor™ Plus 555 (Product # 7400010MP555). Using conjugate solutions: Centrifuge the protein conjugate solution briefly in a microcentrifuge before use; add only the supernatant to the experiment. This step will help eliminate any protein aggregates that may have formed during storage, thereby reducing nonspecific background staining. Because staining protocols vary with application, the appropriate dilution of antibody should be determined empirically.
Applications Tested: This MBSA43 antibody has been tested by flow cytometric analysis of human PBMCs. This may be used at less than or equal to 0.25 µg per test. A test is defined as the amount (µg) of antibody that will stain a cell sample in a final volume of 100 µL. Cell number should be determined empirically but can range from 10^5 to 10^8 cells/test. It is recommended that the antibody be carefully titrated for optimal performance in the assay of interest.

Target Information
TIGIT is a 26 kDa protein that belongs to the poliovirus receptor (PVR) family. It is expressed on NK cells, regulatory T cells, follicular T helper cells, memory CD4+ T cells, and CD8+ T cells, but not on B cells or naive CD4+ T cells. However, TIGIT can be upregulated on naive CD4+ T cells upon activation. TIGIT interacts with members of the PVR and PVR-like families, including CD155. This interaction mediates the engagement of NK and T cells with antigen-presenting cells, fibroblasts, and endothelial cells expressing PVR and PVR-like proteins. Binding of TIGIT to PVR leads to increased secretion of IL-10 and decreased secretion of IL-12B, promoting the generation of mature immunoregulatory dendritic cells and suppressing T cell activation. Through these interactions, TIGIT plays a significant role in modulating immune responses and maintaining immune homeostasis.
