ALOX5 Antibody in Immunohistochemistry (IHC) — Immunohistochemistry analysis of ALOX5 in human lung cancer tissue. Samples were incubated with ALOX5 polyclonal antibody (Product # PA5-95034).

ALOX5 Antibody in Western Blot (WB) — Western blot was performed using Anti-ALOX5 Polyclonal Antibody (Product # PA5-95034), a 70 kDa band corresponding to ALOX5 was observed in THP1 treated with LPS and hTGF beta1 for 72hrs. Expression was also seen in Daudi, Raji cells as compared to K-562, U-937 cells which are reported to be negative for ALOX5 (Figure a); ALOX5 expression was also seen in Mouse, Rat Brain and Spleen in comparison to Mouse Lung, Mouse and Rat Heart and Kidney (Figure b). Whole cell extracts (30 µg lysate) of THP-1 (Lane 1), THP-1 treated with both Lipopolysaccharide (100 ng/mL) and hTGF beta1 (1 ng/mL) for 72hrs (Lane 2), Daudi (Lane 3), Raji (Lane 4), K-562 (Lane 5), U-937 (Lane 6) (Figure a); tissue extracts (30 µg lysate) of Mouse Brain (Lane 1), Mouse Spleen (Lane 2), Mouse Lung (Lane 3), Mouse Heart (Lane 4), Mouse Kidney (Lane 5), Rat Brain (Lane 6), Rat Spleen (Lane 7), Rat Heart (Lane 8), Rat Kidney (Lane 9) (Figure b) were electrophoresed using NuPAGE™ 4-12% Bis-Tris Protein Gel (Product # NP0322BOX). Resolved proteins were then transferred onto a nitrocellulose membrane (Product # IB23001) by iBlot® 2 Dry Blotting System (Product # IB21001). The blot was probed with the primary antibody (0.5 µg/mL) and detected by chemiluminescence with Goat anti-Rabbit IgG (H+L) Superclonal™ Recombinant Secondary Antibody, HRP (Product # A27036, 1:4000 dilution. Uncharacterized band (*) at ~60 kDa in figure a and ~ 40 kDa in figure b was also observed.

Target Information

This gene encodes a protein which, with Arachidonate 5-lipoxygenase-activating protein, is required for leukotriene synthesis. Leukotrienes are arachidonic acid metabolites which have been implicated in various types of inflammatory responses, including asthma, arthritis and psoriasis. This protein localizes to the plasma membrane. Inhibitors of its function impede translocation of 5-lipoxygenase from the cytoplasm to the cell membrane and inhibit 5-lipoxygenase activation.
