Arginase 2 Antibody in Immunocytochemistry (ICC/IF) — Immunocytochemistry-Immunofluorescence analysis of Arginase 2 was performed in HepG2 cells fixed in ice cold MeOH for 5 min. Green: Arginase 2 Polyclonal Antibody (Product # PA5 31088) diluted at 1:500. Blue: Hoechst 33342 staining.

Arginase 2 Antibody in Immunohistochemistry (Paraffin) (IHC (P)) — Arginase 2 Polyclonal Antibody detects ARG2 protein at mitochondria on mouse duodenum by immunohistochemical analysis. Sample: Paraffin-embedded mouse duodenum. Arginase 2 Polyclonal Antibody (Product # PA5-31088) dilution: 1:500. Antigen Retrieval: EDTA based buffer, pH 8.0, 15 min.

Arginase 2 Antibody in Western Blot (WB) — Western blot was performed using Anti-Arginase 2 Polyclonal Antibody (Product # PA5-31088) and a 40 kDa band corresponding to Arginase 2 was observed across cell line and tissues tested. HL-60 showed no expression compared to LNCaP. Whole cell extracts (40 µg lysate) of LNCaP (Lane 1), HL-60 (Lane 2), Mouse liver (Lane 3), Mouse kidney (Lane 4), and Rat kidney (Lane 5) were electrophoresed using Novex® NuPAGE® 4-12 % Bis-Tris gel (Product # NP0321BOX). Resolved proteins were then transferred onto a nitrocellulose membrane (Product # IB23001) by iBlot® 2 Dry Blotting System (Product # IB21001). The blot was probed with the primary antibody (1:1000 dilution) and detected by chemiluminescence with Goat anti-Rabbit IgG (H+L), Superclonal™ Recombinant Secondary Antibody, HRP conjugate (Product # A27036, 1:4000 dilution) using the iBright FL 1000 (Product # A32752). Chemiluminescent detection was performed using Novex® ECL Chemiluminescent Substrate Reagent Kit (Product # WP20005).

Arginase 2 Antibody — Antibody specificity was demonstrated by siRNA mediated knockdown of target protein. LNCaP cells were transfected with Arginase 2 siRNA and reduction of signal was observed in Western Blot using Arginase 2 Polyclonal Antibody (Product # PA5-31088). {KD}

Target Information

Arginase catalyzes the hydrolysis of arginine to ornithine and urea. At least two isoforms of mammalian arginase exists (types I and II) which differ in their tissue distribution, subcellular localization, immunologic crossreactivity and physiologic function. The type II isoform encoded by this gene, is located in the mitochondria and expressed in extra-hepatic tissues, especially kidney. The physiologic role of this isoform is poorly understood; it is thought to play a role in nitric oxide and polyamine metabolism. Transcript variants of the type II gene resulting from the use of alternative polyadenylation sites have been described.
