          Flow cytometry analysis of Axl Recombinant Mouse Monoclonal Antibody (DS7HAXL), Alexa Fluor™ Plus 488 (Product # 750032MP488) was performed using U-87 MG and SH-SY5Y cells. Cells were stained with 0.5 µg of the Axl Recombinant Mouse Monoclonal Antibody (DS7HAXL), Alexa Fluor™ Plus 488 (Product # 750032MP488). Higher expression of Axl was observed in U-87 MG (pink histogram) in comparison to SH-SY5Y (blue histogram).           Alexa Fluor™ Plus recombinant antibodies are conjugated using new, proprietary dye chemistry so you can generate stunning data. Alexa Fluor™ Plus antibodies represent an advancement in fluorescent conjugate technology. Alexa Fluor™ Plus antibodies provide brighter signal compared to leading Alexa Fluor™ antibodies, providing you with better signal-to-noise for your critical experiments. These antibodies show better specificity and lot-to-lot consistency as these are recombinant antibodies, generated by cloning specific genes for the desired antibodies into an expression vector and expressed in vitro.

          Using conjugate solutions: Centrifuge the protein conjugate solution briefly in a microcentrifuge before use; add only the supernatant to the experiment. This step will help eliminate any protein aggregates that may have formed during storage, thereby reducing nonspecific background staining.

          It is recommended that the antibody be carefully titrated for optimal performance in the assay of interest.

          Target Information

          AXL is a chronic myelogenous leukemia-associated oncogene that is also associated with colon cancer and melanoma. The AXL protein is a receptor tyrosine kinase that transduces signals from the extracellular matrix into the cytoplasm by binding growth factors like vitamin K-dependent protein growth-arrest-specific gene 6, and is involved in the stimulation of cell proliferation. TAM receptors are also involved in phagocytosis of apoptotic cells. Mice lacking all three TAM receptors have several degenerative phenotypes linked to inefficient removal of apoptotic cells and membranes (e.g., in the retina and the male reproductive tract) and develop a severe autoimmune phenotype akin to systemic lupus erythematosus, including the production of broad spectrum auto-antibodies. In addition, Axl may function as a putative entry receptor for filoviruses. Axl is also used to identify a specific subpopulation of human blood dendritic cells, also referred to as Siglec 6+/Axl+ dendritic cells. Cellular expression of Axl can be upregulated by TLR ligands, such as LPS or poly I:C. Soluble Axl is generated by proteolytic cleavage of the membrane form. Increased plasma levels may be indicative of inflammation and cancer.
