Store lyophilized/reconstituted at -20°C; once reconstituted make aliquots to avoid repeated freeze-thaw cycles. Please remember to spin the tubes briefly prior to opening them to avoid any losses that might occur from material adhering to the cap or sides of the tube.
For reconstitution add 100 µL of sterile water.

ClpB3 encodes a nucleus-encoded, chloroplast-targeted AAA+ chaperone/disaggregase that is imported into the chloroplast stroma as a processed preprotein with an N-terminal transit peptide; under ambient conditions it is broadly stromal, but during heat stress it can relocalize into discrete stromal foci near the thylakoid membrane system, consistent with a role at sites of aggregate deposition. Structurally, ClpB3 is a class I Hsp100/Clp chaperone that assembles into an ATP-dependent homohexameric ring and contains the characteristic N-terminal domain, an inserted coiled-coil middle domain, and two tandem AAA+ ATPase modules with Walker-type nucleotide-binding motifs that power substrate engagement and threading through a central pore. Functionally, ClpB3 supports chloroplast proteostasis by recognizing and resolubilizing stress-denatured and aggregated proteins using ATP hydrolysis, thereby promoting recovery of chloroplast protein homeostasis after heat stress; genetic and biochemical data indicate it is required in vivo for efficient clearance of heat-induced chloroplast protein aggregates and contributes to cellular thermotolerance, and purified plant ClpB3 can directly reactivate aggregated model substrates in an ATP-dependent manner.
