Cyclin D3 Antibody in Western Blot (WB) — Western blot analysis of Cyclin D3 in lane 1: rat testis tissue lysate, lane 2: rat thymus tissue lysate, lane 3: rat lung tissue lysate, lane 4: rat ovary tissue lysate, lane 5: JURKAT whole cell lysate, lane 6: A549 whole cell lysate, lane 7: MCF-7 whole cell lysate, lane 8: HeLa whole cell lysate using 50 µg (reducing conditions) per well. Electrophoresis was performed on 5-20% SDS-PAGE gel at 70V (Stacking gel) / 90V (Resolving gel) for 2-3 hours and protein was transferred to a nitrocellulose membrane at 150mA for 50-90 minutes. Sample was blocked with 5% Non-fat Milk/TBS for 1.5 hours at room temperature, incubated with Cyclin D3 polyclonal antibody (Product # PA5-78943) at a dilution of 0.5 µg/mL (overnight at 4°C), followed by goat anti-rabbit IgG-HRP secondary antibody at a dilution of 1:10,000. Signal development was performed using a chemiluminescence (ECL) kit.

Target Information

The immunogen for the anti-h-Cyclin D3 was a recombinant full length Cyclin D3 protein recognizing a 34 kD protein. Cyclin D3 is a G1 cyclin closely related to Cyclins D1 and D2 phase progression. Like Cyclin D1 and Cyclin D2, it is activated by the same mechanisms and can phosphorylate pRB when associated with cdk4 and cdk6.
