EBI3 is a 228 amino acid containing pro- and anti-inflammatory cytokine belonging to the type I cytokine receptor family and contains two fibronectin type-III domain. It forms a heterodimer with IL-27 and is also a component of the IL-12 heterodimer. A protein involved in regulated cell-mediated immune responses, EBI3 has an important role in regulating T helper cell development, suppressing T-cell proliferation, stimulating cytotoxic T-cell activity, inducing isotype switching in B-cells, and has diverse effects on innate immune cells. Target cells of EBI3 include CD4 T helper cells, type 1 effector cells (TH1), type 2 effector cells (TH2) and IL-17 producing helper T-cells (TH17). It drives rapid clonal expansion of naive but not memory CD4 T-cells. EBI3 is important in anti-tumor as well as anti-angiogenic activity with activation and production of anti-angiogenic chemokines. It also strongly synergizes with IL-12 to trigger interferon-gamma/IFN-gamma production of naive CD4 T-cells.
