Target Information
GAI encodes a nuclear DELLA-class GRAS regulatory protein that functions as a central repressor within gibberellin (GA) signaling. The polypeptide is organized with an amino-terminal regulatory region containing the conserved DELLA and adjacent VHYNP and LExLE motifs that mediate GA-dependent binding to the soluble GA receptor GID1, and a carboxy-terminal GRAS domain typical of plant transcriptional regulators that supports protein-protein interactions and transcriptional control. In the presence of bioactive GA, GID1 engages the GAI DELLA region to form a hormone-dependent complex that promotes relief of DELLA-mediated growth restraint, commonly through destabilization and turnover via the ubiquitin-26S proteasome pathway; conversely, GA-insensitive variants with lesions in the DELLA region fail to interact productively with GID1 and remain active repressors. Through this receptor-coupled "derepression" mechanism, GAI limits GA-responsive gene expression and restrains cell proliferation/expansion, thereby modulating major developmental outputs such as stem elongation, vegetative growth, and timing of floral initiation.
