Description: Wnt proteins constitute a family of secreted glyocoproteins that activate signal transduction pathways to mediate tissue homeostasis, cell fate, cell proliferation, and self-renewal. The canonical signaling pathway is initiated upon Wnt binding to a Frizzled family receptor and an LRP5/LRP6 coreceptor, which leads to activation of the beta-catenin cascade. Non-canonical Wnt signals are activated by binding to a Frizzled family receptor and ROR2 coreceptor. Extensive research has shown that deregulation of these pathways or its components can lead to cancer.
Human Wnt-7a is encoded as a 349 amino acid precursor that, upon secretion, is cleaved into a 318 amino acid (48 kDa) mature form. At the amino acid level, the mature form of human Wnt-7a is approximately 98% identical to mature mouse Wnt-7a. Depending on the tissue, Wnt-7a can activate the canonical and non-canonical signaling pathways. Studies have demonstrated its involvement in a variety of functions, including neurogenesis, muscle regeneration, and carcinogenesis.
Applications Reported: Recombinant human Wnt-7a is biologically active and can inhibit Wnt-3a-induced alkaline phosphatase production in MC3T3-E1 cells.
Applications Tested: The ED50 of this protein, as determined by Wnt-3a-induced alkaline phosphatase production in MC3T3-E1 cells, is 40-60 ng/mL.

Storage and handling: For best recovery, quick spin vial prior to opening. Reconstitute in water to a concentration of less than or equal to 0.2 mg/mL. Do not vortex. Use in sterile environment. Store lyophilized protein at less than or equal to -20°C. Reconstituted protein solution can be stored with carrier-protein (e.g., 0.1% BSA) at less than or equal to -20°C.

Studies have demonstrated its involvement in a variety of functions, including embryonic development and carcinogenesis. Wnt-1 signaling can also induce the phosphorylation and subsequent degradation of c-Myb.
