The target sequence has 98% sequence homology with mouse, rat and bovine, and 91% sequence homology with xenopus and zebrafish.
Jumonji domain containing 6 is a histone demethylating enzyme, which is active on arginine residues, but not lysines. Its activity destabilizes heterochromatin structures and activates gene expressions in euchromatin. In endothelial cells, the activity of JMJD6 is necessary for angiogenesis. If JMJD6 expression is reduced, the VEGF-receptor (Flt-1) is incorrectly spliced, leading to increased soluble Flt-1 which binds VEGF and PlGF and inhibits angiogenesis. JMJD6 also hydroxylates RNA splicing factors, and is important in embryonic differentiation. It most likely interacts specifically with single-stranded RNA, which may contribute to its early differentiation importance.
