LNX2 (Ligand of Numb Protein X 2) protein is encoded by the LNX2 gene located on chromosome 13 in humans and is known as PDZRN1 (PDZ domain-containing RING finger 1). It is through its PDZ domains that LNX2 forms oligomers and interacts with some of its target proteins. LNX2 is best studied as a E3 ubiquitin ligase that also contains the NPXY motifs which allows binding to the Numb protein. This interaction has been shown to be responsible for ubiquitination of Numb, which prevents the proteasomal degradation of Notch intracellular domain (NICD) and thus modulates the Notch signaling pathway.
