HCN4 Antibody (MA5-45433) in ICC/IF
Immunocytochemistry/Immunofluorescence analysis using human neuroblastoma cells. Fixation involved 4% PFA for 15 min. Samples were incubated with HCN4 monoclonal antibody (Product # MA5-45433) at 1:100 for overnight at 4°C with slow rocking, followed by AlexaFluor 488 at 1:1,000 for 1 hour at RT. Counterstain used was Phalloidin-iFluor 647 (red) F-Actin stain; Hoechst (blue) nuclear stain at 1:800, 1.6mM for 20 min at RT. (A) Hoechst (blue) nuclear stain. 1 µg/mL of MA5-45433 was sufficient for detection of HCN4 in 10 µg of COS cell lysate transiently transfected with HCN4 by colorimetric immunoblot analysis using Goat anti-mouse IgG:HRP as the secondary antibody.|Detects approximately 130kDa. No cross-reactivity against other HCNs.
Target Information
HCN4 is a member of the family of hyperpolarization-activated and cyclic-nucleotide-gated (HCN) channels. HCN currents have been linked to pacemaker activity in the heart and brain, resting potential control, as well as neuronal plasticity. It has been shown that HCN4 channels function as receptors for sour taste, and are associated with pacemaker potential generation in the sinoatrial node.
