Target Information

TAOK kinases, including TAOK1, TAOK2, and TAOK3, are serine-threonine kinases that belong to the sterile 20 (STE20) family and play significant roles in various cellular processes, including proliferation, apoptosis, and tumor progression. TAOK1 is implicated in neurodevelopmental disorders and has an evolving phenotype and genotype spectrum, which includes associations with macrocephaly and autism spectrum disorders. The TAOK2 gene, located on chromosome 16, is a crucial kinase participating in the regulation of protein translation through the phosphorylation of eukaryotic elongation factor 2 (eEF2) and is also involved in neurodevelopmental pathways. Moreover, TAOK2 acts as a potential tumor suppressor, with its downregulation linked to cancer progression in numerous types of cancer. TAOK3, another member of this kinase family, is expressed ubiquitously and has been linked to the regulation of age-associated inflammation, acting through pathways that limit pro-inflammatory monocyte accumulation. Additionally, TAOK3 is involved in osteoblast differentiation and modulates T cell receptor signaling by influencing components like SHP-1 and LCK. All three kinases are crucial for maintaining cellular homeostasis and their aberrations can lead to pathological conditions, particularly related to neurodevelopment and cancer.
