PIM1 Antibody in Western Blot (WB) — Western blot analysis of K562 cell lysates using Zymed Ms anti-Pim-1 (Product # 39-4600).

PIM1 Antibody in ChIP Assay (ChIP) — Chromatin immunoprecipitation analysis of PIM-1 was performed using cross-linked chromatin from Human J9.2 J-lat T lymphocytes culture treated with 10 µg/mL PHA (phytohemaglutin) for 0, 2, and 24 hours. Immunoprecipitation was performed using a multiplex microplate Matrix ChIP assay (see reference for Matrix ChIP protocol: http://www.ncbi.nlm.nih.gov/pubmed/22098709) with 1.0 µL/100 µL well volume of a PIM-1 monoclonal antibody (Product # 39-4600). Chromatin aliquots from ~1 x 104 cells were used per ChIP pull-down. Quantitative PCR data were done in quadruplicate using 1 µL of eluted DNA in 2 µL SYBR real-time PCR reactions containing primers shown to amplify -15kb and exon-1 of the EGR1 gene. PCR calibration curves were generated for each primer pair from a dilution series of sheared total genomic DNA. Quantitation of immunoprecipitated chromatin is presented as signal relative to the total amount of input chromatin. Results represent the mean +/- SEM for three experiments. PIM1 Antibody — The specificity of anti-PIM1 monoclonal antibody (Product # 39-4600) was demonstrated using chromatin immunoprecipitation (ChIP). q-PCR was used to demonstrate levels of enrichment of PIM1 at the expected loci in exon 1 of human EGR1 gene but not negative site in -15kb region of the same gene in Human J9.2 J-lat T lymphocytes culture treated with 10 µg/mL PHA (phytohemaglutin) for 2 hours compared to those in untreated cells. {TM}

Target Information

PIM1, which belongs to the Serine/Threonine protein kinase family, is thought to play a role in signal transduction in blood cells. The protooncogene PIM1 encodes a protein kinase upregulated in prostate cancer. It may affect the structure or silencing of chromatin by phosphorylating HP1 gamma/CBX3. PIM1 is expressed primarily in cells of the hematopoietic and germ line lineages.
