          PUMA alpha Antibody (PA5-20007) in ICC/IF

          Immunocytochemistry of K562 cells using PUMA Polyclonal Antibody (Product # PA5-20007) at 1 µg/mL. Cells were fixed with formaldehyde and blocked with 0.1 serum for 1 h at RT; antigen retrieval was by heat mediation with a citrate buffer (pH6). Samples were incubated with primary antibody overnight at 4°C. A goat anti-rabbit IgG H&L (HRP) at 1:250 was used as secondary.           Product Specific Information

          The PA5-20007 immunogen is located within the last 50 amino acids of PUMA. The immunogenic sequence is identical between the A and B forms of PUMA.

          In Western blot applications, this antibody detects a band at ~23kDa. A suggested positive control is K562 cell lysate.

          PA5-20007 can be used with blocking peptide PEP-0127.

          Predicted species reactivity based on immunogen sequence: Rat: (100%), Mouse: (100%)

          Target Information

          PUMA is one of the pro-apoptotic Bcl-2 family members including Bax and Noxa, which are also transcriptional targets of p53. The PUMA gene encodes two BH3 domain-containing proteins termed PUMA-a and PUMA-b. PUMA proteins bind Bcl-2, localize to the mitochondria, and induce cytochrome c release and apoptosis in response to p53. PUMA may be a direct mediator of p53-induced apoptosis. Antisense inhibition of PUMA expression reduced the apoptotic response to p53, and PUMA is likely to play a role in mediating p53-induced cell death through the cytochrome c/Apaf-1-dependent pathway.
