Western blot analysis of Prothrombin in extracts of HepG2 cells. Samples were incubated with Prothrombin Polyclonal antibody (Product # PA5-120155) using a dilution of 1:500, followed by HRP Goat Anti-Rabbit IgG (H+L) at a dilution of 1:10,000. Lysates/proteins: 25 µg per lane. Blocking buffer: 3% nonfat dry milk in TBST. Detection: ECL Basic Kit. Exposure time: 180s.

Target Information
Prothrombin or Coagulation factor II, is proteolytically cleaved to form thrombin in the first step of the coagulation cascade which ultimately results in the stemming of blood loss. F2 also plays a role in maintaining vascular integrity during development and postnatal life. Mutations in F2 leads to various forms of thrombosis and dysprothrombinemia.
