Store lyophilized/reconstituted at -20°C; once reconstituted make aliquots to avoid repeated freeze-thaw cycles. Please remember to spin the tubes briefly prior to opening them to avoid any losses that might occur from material adhering to the cap or sides of the tube.

RPB1 encodes the largest catalytic subunit of eukaryotic RNA polymerase II (the POLR2A gene product), a predominantly nuclear, chromatin-associated enzyme complex responsible for DNA-templated transcription of protein-coding genes into precursor mRNA. The RPB1 polypeptide forms the core of the polymerase active-center framework and, together with other polymerase subunits, contributes to the DNA-binding cleft that positions the template strand for RNA synthesis. A defining structural feature is its carboxy-terminal domain (CTD), a low-complexity tail composed of tandem heptapeptide repeats with the consensus sequence YSPTSPS; in mammals the CTD comprises 52 repeats and is essential for polymerase activity. Dynamic post-translational modification of CTD residues-especially phosphorylation of Ser2 and Ser5 across the repeats-coordinates transitions through initiation, elongation, and termination and functionally couples transcription to co-transcriptional RNA processing by recruiting and organizing factors required for 5-prime capping, splicing, and 3-prime end cleavage/polyadenylation on nascent transcripts.
