SATB1 Antibody in Western Blot (WB) — Knockdown of SATB1 was achieved by transfecting IMR-32 with SATB1 specific siRNAs (Silencer® select Product # s12481, s12479). Western blot analysis (Fig. a) was performed using whole cell extracts from the SATB1 knockdown cells (Lane 3), non-specific scrambled siRNA transfected cells (Lane 2) and untransfected cells (Lane 1). The blot was probed with SATB1 Polyclonal Antibody (Product # PA5-19148, 0.3ug/ml) and Rabbit anti-Goat IgG (H+L) Superclonal™ Recombinant Secondary Antibody, HRP (Product # A27014, 1:4000 dilution). Densitometric analysis of this western blot is shown in histogram (Fig. b). Decrease in signal upon siRNA mediated knock down confirms that antibody is specific to SATB1.

SATB1 Antibody in Immunohistochemistry (Paraffin) (IHC (P)) — Immunohistochemical analysis of SATB1 in paraffin embedded human thymus using a SATB1 polyclonal antibody (Product #PA5-19148) at a concentration of 3.8 µg/mL. Steamed antigen retrieval was performed with pH 6 buffered citrate. Samples were then stained with alkaline phosphatase.

SATB1 Antibody in Immunofluorescence (IF) — Immunofluorescence analysis of HUR was performed using 70% confluent log phase IMR-32 cells. The cells were fixed with 4% paraformaldehyde for 10 minutes, permeabilized with 0.1% Triton™ X-100 for 15 minutes, and blocked with 2% BSA for 1 hour at room temperature. The cells were labeled with HUR Rabbit Polyclonal Antibody (Product # PA5-19148) at 5ug/ml in 0.1% BSA, incubated at 4 degree celsius overnight and then labeled with Rabbit anti-Goat IgG (H+L) Cross-Adsorbed Secondary Antibody, Alexa Fluor 488 (Product # A-11078) at a dilution of 1:2000 for 45 minutes at room temperature (Panel a: green). Nuclei (Panel b: blue) were stained with ProLong™ Diamond Antifade Mountant with DAPI (Product # P36962). F-actin (Panel c: red) was stained with Rhodamine Phalloidin (Product # R415). Panel d represents the merged image showing Nuclear localization. Panel e represents control cells with no primary antibody to assess background. The images were captured at 60X magnification.

SATB1 Antibody in Knockdown — Antibody specificity was demonstrated by siRNA mediated knockdown of target protein. IMR-32 cells were transfected with SATB1 siRNA and a decrease in signal intensity was observed in western blot application using Anti-SATB1 Polyclonal Antibody (Product # PA5-19148). {KD}

SATB1 Antibody in Relative expression — Antibody specificity was demonstrated by detection of differential basal expression of the target across tissues tested owing to their inherent genetic constitution. Relative expression of SATB1 was observed in Mouse and Rat Thymus in comparison to Mouse and Rat Kidney using Anti-SATB1 Polyclonal antibody (Product # PA5-19148) in Western Blot. An uncharacterized band (*) of ~60 kDa was also observed in Mouse Thymus tissue.. {RE}

Target Information

SATB1 (DNA-binding protein SATB1) is a crucial silencing factor that contributes to the initiation of X inactiviation mediated by Xist RNA that occurs during embryogenesis and in lymphoma. SATB1 binds to DNA at special AT-rich sequences, the consensus SATB1-binding sequence, and at nuclear matrix- or scaffold-associated regions. It is thought to recognize the sugar-phosphate structure of double-stranded DNA. SATB1 is also a transcriptonal repressor controlling nuclear and viral gene expression in a phosphorylated and acetylated status-dependent manner by binding to matrix attachment regions of DNA and inducing a local chromatin-loop remodeling. SATB1 modulates the genes that are essential in the maturatio of the immune T-cell CD8SP from thymocytes. Alu-like motifs and SATB1-binding sites provide an unique chromatin context which seems preferentially targeted by the HIV-1 integration machinery. Moreover, HIV-1 Tat may overcome SATB1-mediated repression of IL1 and IL2RA in T-cells by binding to the same domain as HDAC1.
