SEMA4C Antibody in Immunohistochemistry (Paraffin) (IHC (P)) — Immunohistochemical analysis of Semaphorin 4C was detected in immersion fixed paraffin-embedded sections of human brain (cortex) using Sheep Anti-human Semaphorin 4C Antigen Affinity-purified Polyclonal Antibody (Product # PA5-47814) at 15 µg/mL overnight at 4 °C. Before incubation with the primary antibody, tissue was subjected to heat-induced epitope retrieval. Tissue was stained using the Anti-Sheep HRP-DAB (brown and counterstained with hematoxylin (blue). Specific staining was localized to neuronal cell bodies.

SEMA4C Antibody in Western Blot (WB) — Western blot analysis from lysates of SW13 human adrenal cortex adenocarcinoma cell line. PVDF membrane was probed with 0.5 µg/mL of Sheep Anti-human Semaphorin 4C Antigen Affinity-purified Polyclonal Antibody (Product # PA5-47814) followed by HRP-conjugated Anti-Sheep IgG Secondary Antibody. A specific band was detected for Semaphorin 4C at approximately 110 kDa (as indicated). This experiment was conducted under reducing conditions.

In direct ELISAs, approximately 50% cross-reactivity with recombinant mouse (rm) Sema4C is observed, and approximately 10% cross-reactivity with recombinant human (rh) Sema4G is observed, and less than 5% cross-reactivity with rhSema3B and rmSema3F is observed.

Reconstitute at 0.2 mg/mL in sterile PBS.

Target Information

SEMA4C (semaphorin-4C), also known as SEMAF, is an 833 amino acid single-pass type I membrane protein that contains one sema domain, one PSI domain and one Ig-like C2-type domain. Expressed in a variety of tissues, including lung, kidney and brain, SEMA4C interacts with GIPC and NCDN and is thought to play a role in the formation of neural networks during development.
