The Toll-like receptor (TLR) family in mammal comprises a family of transmembrane proteins characterized by multiple copies of leucine rich repeats in the extracellular domain and IL-1 receptor motif in the cytoplasmic domain. Like its counterparts in Drosophila, TLRs signal through adaptor molecules (1). TIR8, also know as single Ig IL-1 receptor, is a member of the IL-1/TLR superfamily (2). TIR8 acts as intracellular decoy to inhibit NF-kappaB activation by members of the IL-1/TLR family (3, 4).
