SNRPE Antibody in Immunocytochemistry (ICC/IF) — Immunofluorescence analysis of SNRPE was performed using 70% confluent log phase HEK-293 cells. The cells were fixed with 4% paraformaldehyde for 10 minutes, permeabilized with 0.1% Triton™ X-100 for 10 minutes, and blocked with 2% BSA for 45 minutes at room temperature. The cells were labeled with SNRPE Polyclonal Antibody (Product # PA5-78173) at 1:100 in 0.1% BSA, incubated at 4 degree celsius overnight and then labeled with Goat anti-Rabbit IgG (H+L) Superclonal™ Recombinant Secondary Antibody, Alexa Fluor® 488 conjugate (Product # A27034), (1:2000), for 45 minutes at room temperature (Panel a: Green). Nuclei (Panel b:Blue) were stained with SlowFade® Gold Antifade Mountant with DAPI (Product # S36938). F-actin (Panel c: Red) was stained with Rhodamine Phalloidin (Product # R415, 1:300). Panel d represents the merged image showing cytoplasm and nuclear localization. Panel e represents control cells with no primary antibody to assess background. The images were captured at 60X magnification.

SNRPE Antibody in Immunohistochemistry (Paraffin) (IHC (P)) — SNRPE Polyclonal Antibody detects SNRPE protein at nucleus by immunohistochemical analysis. Sample: Paraffin-embedded mouse brain. SNRPE stained by SNRPE Polyclonal Antibody (Product # PA5-78173) diluted at 1:500. Antigen Retrieval: Citrate buffer, pH 6.0, 15 min.

SNRPE Antibody in Western Blot (WB) — Western blot was performed using Anti-SNRPE Polyclonal Antibody(Product # PA5-78173) and a 11kDa band corresponding to SNRPE was observed across the cell lines tested. Whole cell extracts (30µg lysate) of HEK-293 (Lane 1), K-562 (Lane 2), Hep G2 (Lane 3), HeLa (Lane 4) were electrophoresed using Novex™ 16% Tricine Protein Gel (Product # EC6695BOX). Resolved proteins were then transferred onto a Nitrocellulose membrane (Product # LC2001) by iBlot® 2 Dry Blotting System (Product # IB21001). The blot was probed with the primary antibody (1:1000 dilution) and detected by chemiluminescence with Goat anti-Rabbit IgG (H+L) Superclonal™ Recombinant Secondary Antibody, HRP (Product # A27036,1:4000 dilution) using the iBright FL 1000 (Product # A32752). Chemiluminescent detection was performed using Novex® ECL Chemiluminescent Substrate Reagent Kit (Product # WP20005).

SNRPE Antibody — Antibody specificity was demonstrated by siRNA mediated knockdown of target protein. HEK-293 cells were transfected with SNRPE siRNA and decrease in signal intensity was observed in Western Blot application using Anti-SNRPE Polyclonal Antibody (Product # PA5-78173). {KD}

Target Information

The protein encoded by SNRPE is a core component of U small nuclear ribonucleoproteins, which are key components of the pre-mRNA processing spliceosome. The encoded protein plays a role in the 3' end processing of histone transcripts. This protein is one of the targets in the autoimmune disease systemic lupus erythematosus, and mutations in SNRPE have been associated with hypotrichosis. Several pseudogenes of SNRPE have been identified.
