Cryo-electron tomography: structural biology on a proteomic scale

Cryo-electron tomography (cryo-ET) enables three-dimensional visualization of cells, organelles and macromolecular complexes in their near-native state. By combining cryogenic preservation with tilt-series acquisition and computational reconstruction, cryo-ET bridges cellular context with molecular resolution, revealing biological architecture as it exists in situ.

 

As the field advances, cryo-ET is evolving from highly specialized experiments toward more automated, repeatable and higher-throughput structural investigations. Innovations in cryo-focused ion beam (cryo-FIB) milling of cells and tissues, increasingly streamlined tomography data acquisition, and faster, more sophisticated image processing workflows are helping to improve reproducibility, efficiency and accessibility.
