Immunofluorescence analysis of VAV1 was performed using 70 confluent log phase Jurkat cells. The cells were fixed with 4% paraformaldehyde for 10 minutes, permeabilized with 0.1% Triton™ X-100 for 15 minutes, and blocked with 2% BSA for 45 minutes at room temperature. The VAV1 gene, located on chromosome 19p13.2, encodes for a protein that functions as a guanine nucleotide exchange factor (GEF). Structurally, the VAV1 protein contains several functional domains, including Dbl homology (DH) and pleckstrin homology (PH) domains, which facilitate its role in exchange activity, and SH3 and SH2 domains, critical for signal transduction. VAV1 is primarily expressed in hematopoietic cells and plays a crucial role in the regulation of immune responses, particularly in T-cell receptor signaling.
