Applied Biosystems Custom TaqMan Gene Expression assays are the custom version of our predesigned TaqMan assays for quantitative real-time PCR. The Custom TaqMan Gene Expression assays consist of a pair of unlabeled PCR primers and a TaqMan probe with a dye label (FAM, VIC, ABY, JUN, and Cyanine 5) on the 5' end and a minor groove binder (MGB) and nonfluorescent quencher (NFQ) on the 3' end.

Can I delete an assay or sample from my qPCR study?

An assay or sample may be deleted from a study only if there is no data or wells associated with it. Upon import of an experiment, the software collects all the assays and samples from the plate and lists them in the Setup > Assays or Setup > Samples workspaces. The assays and samples are stored in these workspaces as a library, and remain there even if you delete the experiment from the Study. Deleting the experiment will remove any data (wells) associated with the assays or samples, but not the assays or samples from the library. The assays and samples must then be deleted from these workspaces to remove them from the Study.

How do I determine if Applied Biosystems TaqMan Assays that I want to multiplex are compatible with each other?

You can utilize the Applied Biosystems TaqMan Multiplex Interaction Check (MIC) tool. The MIC tool uses bioinformatics to identify potential interactions between primers, probes, and amplicons that could negatively impact multiplex qPCR performance. The tool was developed and validated using data from approximately 200 multiplex panels, combining computational analysis with wet-lab testing. By flagging assay combinations that may be prone to false positives or reduced sensitivity, MIC tool helps researchers design multiplex panels with greater confidence and reduce time spent on trial-and-error optimization.risk and observed multiplex performance.

Why are the concentrations of forward and reverse primer in our Primer-Limited (PL) Applied Biosystems TaqMan Gene Expression Assays?

Primer-Limited Applied Biosystems TaqMan Gene Expression Assays with FAM, ABY, JUN, or Cyanine 5 dyes have a primer concentration of 350 nM. Assays with VIC dyes have a primer concentration of 150 nM.

Why are some Applied Biosystems TaqMan Gene Expression Assays only available in Primer-Limited concentrations?

In a multiplex qPCR reaction, like in Applied Biosystems TaqMan Gene Expression Assays, targets often differ in abundance and amplification efficiency. Highly expressed or highly efficient targets can consume a disproportionate share of reaction components, generate very strong fluorescent signals, and outcompete weaker targets for polymerase and other reagents. Primer-limited assays provide balanced amplification of targets in multiplex workflows by reducing primer competition between assays and supporting more consistent amplification across multiple targets.
