We haven't tested this extensively, but we have tested storage of the cDNA and yellow sample buffer premixed for 2-3 days. For best practice, we recommend keeping the cDNA and yellow sample buffer separate until the reaction set up.

The incubation time is not required as UNG is active at room temperature and will degrade amplicons with dUs present.
