The Thermo Scientific™ SureTect™ Escherichia coli STEC Identification PCR Assay is part of a complete workflow for the rapid detection of Escherichia coli O157:H7, stx, eae, and the Escherichia coli STEC serotypes (O26, O45, O103, O111, O121, O145) from food samples. The assay incorporates an optimized liquid lysis step, providing a quick and simple method for preparing the sample for rapid detection on the Applied Biosystems™ QuantStudio™ 5 Food Safety Real-Time PCR System and software.
Samples assessed as positive using the SureTect™ Escherichia coli O157:H7 and STEC Screening PCR Assay are further tested using this SureTect™ Escherichia coli STEC Identification PCR Assay. The assay has been AOAC-RI and AFNOR (ISO 16140-2:2016) validated on a range of food, including low and high sample sizes of raw meat (excluding poultry), vegetables, fruits, juices, and dairy samples, and it enables users to reliably detect and differentiate Shiga-toxin producing Escherichia coli O157:H7 and other STEC in as little as 10 hours.

The lysate of screened positive samples is pipetted into the Escherichia coli STEC Identification PCR tubes to re-hydrate the pre-loaded PCR pellet which contains all the necessary components and reagents for PCR, including a probe, primers and DNA template for the internal amplification control (IAC). The PCR tubes are sealed, then loaded into the Applied Biosystems™ QuantStudio™ 5 Food Safety Real-Time PCR instrument using RapidFinder™ Analysis Software v3.0 or later. After completing the run, interpreted results are clearly displayed and can be reported, stored, printed or downloaded as required. The internal amplification control confirms correct assay conditions have occurred.
