5-CFDA, AM ist ein membrangängiges Esterasesubstrat, das als Viabilitätssonde dienen kann, die sowohl die zur Aktivierung seiner Fluoreszenz erforderliche enzymatische Aktivität als auch die für die intrazelluläre Retention ihres Fluoreszenzprodukts erforderliche Zellmembranintegrität misst. Elektronisch neutrale AM-Ester von CFDA können zur Aufnahme in Zellen in geringeren Konzentrationen als CFDA eingesetzt werden. Nach der Hydrolyse durch intrazelluläre Esterasen liefert dieser AM-Ester auch Carboxyfluorescein: Im Vergleich zu Fluorescein enthält Carboxyfluorescein zusätzliche negative Ladungen und wird daher besser in Zellen aufbewahrt.
