Description

The Human α-Synuclein ELISA research-use-only kit is designed to detect and quantify the level of human α-Synuclein protein using 96-well plates and a microplate reader in cell lysates or tissue homogenates independent of its phosphorylation state. Although performance characterization of this ELISA kit was done primarily on human cell lines, cross-reactivity of this kit with mouse and rat cells was not observed.

Principle of the method
The Invitrogen™ α-Synuclein kit is a solid phase sandwich enzyme-linked immunosorbent assay (ELISA). A monoclonal antibody specific for α-synuclein has been coated onto the wells of the microtiter strips provided. Samples, including a standard containing α-synuclein, control specimens, and unknowns, are pipetted into these wells. During the first incubation, the α-synuclein antigen binds simultaneously to the immobilized (capture) antibody and to the solution phase rabbit polyclonal (detection) antibody specific for α-synuclein. After washing, a horseradish peroxidase–labeled anti-rabbit IgG (anti-rabbit IgG-HRP) is added. This binds to the detection antibody to complete the four-member sandwich. After the second incubation and washing to remove all the excess anti-rabbit IgG-HRP, a substrate solution is added, which is acted upon by the bound enzyme to produce color. The intensity of this colored product is directly proportional to the concentration of α-synuclein present in the original specimen.

Background information
α-Synuclein (accession number NM_000345; known alternatively as NACP, PARK1, and PD1 [Parkinson Disease, familial 1]) is a member of the synuclein family of proteins that also includes β-Synuclein and γ-Synuclein. α-synuclein is highly expressed in several areas of the brain, including the substantia nigra, thalamus, hippocampus, amygdala, corpus callosum, and caudate nucleus, appearing both in neurons where it concentrates at synapses, and in glial cells. Most other tissues, with the exception of liver, express α-synuclein at relatively low levels. Several physiological roles for α-synuclein have been suggested. This protein appears to play a role in dopamine metabolism and vesicle trafficking. It also appears to regulate membrane permeability.
