Cell adhesion molecule 3 (CADM3), also known as Necl-1, is a gene located on chromosome 1p36.22, encoding a protein that is part of the immunoglobulin superfamily involved in cell-cell adhesion processes. CADM3 is predominantly expressed in the nervous system, particularly in regions such as the brain and spinal cord, where it plays crucial roles in neuronal communication, synapse formation, and maintenance of neural network architecture. The protein functions by mediating homophilic and heterophilic interactions with other cell adhesion molecules, facilitating synapse stabilization and neuronal differentiation. CADM3's activity is essential for proper neural development and function, and dysregulation of its expression has been linked to various neurological disorders. Beyond its role in the nervous system, CADM3 also contributes to tumor suppression in certain cancers, as its expression is often downregulated in tumor tissues. Research into CADM3 continues to explore its potential as a therapeutic target in both neurobiology and oncology.
