Pro-IL-33 is the precursor form of interleukin-33 (IL-33), a cytokine that plays an essential role in the immune system, particularly in the regulation of inflammatory and immune responses. IL-33 is a member of the IL-1 family and is produced by various cell types, including epithelial cells, endothelial cells, and fibroblasts. In its precursor form, pro-IL-33 is biologically active and can be cleaved by caspases or other proteases to generate the mature, active cytokine. IL-33 functions by binding to its receptor, ST2 (IL1RL1), on the surface of target cells, which include a variety of immune cells such as Th2 cells, mast cells, and group 2 innate lymphoid cells (ILC2s). This interaction leads to the activation of signaling pathways that promote the production of Th2-associated cytokines, contributing to the immune response against infections and playing a role in allergic inflammation. Elevated levels of IL-33 have been associated with various inflammatory and autoimmune diseases, including asthma, rheumatoid arthritis, and inflammatory bowel disease (IBD). Due to its significant role in modulating immune responses, pro-IL-33 is a critical target for research aimed at developing new treatments for inflammatory and autoimmune conditions.
