Yes. Once reconstituted, pHrodo BioParticles Conjugates for Phagocytosis and Phagocytosis Kit, for Flow Cytometry (Cat. Nos. P35367, P35361, P35360, P35366, P35364, P35365, A10010) can be stored at 2 - 8 degrees C for several weeks, as long as sodium azide is added to a final concentration of 2 mM. If no sodium azide is added, the cell suspension needs to be used right away or on the same day to avoid contamination. DO NOT FREEZE the resuspended pHrodo bioparticle conjugates.

While the bacteria have been attenuated with formaldehyde and alcohol desiccation, the BioParticles products are not considered sterile, and we do not recommend incubation of more than 4 hours. This applies to all of our dye-labeled (pHrodo, Alexa Fluor, etc.) and unlabeled BioParticles products.

Yes. You can use cell types for pHrodo Deep Red E. Coli Bioparticles Conjugate for Phagocytosis (Cat. No. 35360) other than RAW or MMM (J774A.1). The final concentration of BioParticles per cell and incubation time would have to be optimized if using other cell types. You may follow the protocol for the RAW and MMM cells and determine if any changes are required for your cells of interest.

The limitations in using pHrodo Deep Red E. coli BioParticles Conjugate for Phagocytosis (Cat. No. P35360) are with autofluorescence from the cells or surrounding material that can interfere with the signal from pHrodo Deep Red E. coli Bioparticles. Before use, examine unlabeled, unstained samples for any autofluorescence in the far-red range. Some natural pigments and compounds accumulated by the cells may fluoresce in the far-red emission range.

The pHrodo Phagocytosis Kits that specifically include "for Flow Cytometry" in the product name were optimized and packaged specifically for use with whole blood samples.The "for Flow Cytometry" kits include buffers that are not provided in the other kits. For the pHrodo Phagocytosis Kits that do not include "for Flow Cytometry" in the product title can also be used for flow cytometry assays, but you must provide all necessary buffers and optimize all assay parameters.
