          RAR gamma-1 Antibody (PA3-813) in WB

          Western blot was performed using Anti-RAR gamma-1 Polyclonal Antibody (Product # PA3-813) and a 60 kDa band corresponding to RAR gamma-1 was observed in MCF-7, HaCaT and Mouse Mammary Gland but not in BeWo and HepG2. Nuclear enriched extracts (50 µg lysate) of MCF7 (Lane 1), BeWo (Lane 2), HaCaT (Lane 3), Hep G2 (Lane 4) and tissue extract (50 µg lysate) of Mouse Mammary gland (Lane 5) were electrophoresed using NuPAGE™ 10% Bis-Tris Protein Gel (Product # NP0301BOX).           Product Specific Information

          PA3-813 detects recombinant human retinoic acid receptor (RAR) gamma-1. The immunizing sequence is 100% conserved in mouse. This antibody detects RAR gamma-1, but not RAR gamma-2, or either of the alpha or beta forms of RAR.

          PA3-813 has been successfully used in Western blot, immunoprecipitation and gel shift procedures. By Western blot, this antibody detects an ~50 kDa protein representing RAR gamma-1 from transiently transfected 293T cells. PA3-813 has been shown to efficiently super shift the human RAR gamma-1/retinoid X receptor (RXR)/RAR response element complex in gel super shift experiments using a number of different response elements.

          PA3-813 immunizing peptide corresponds to the N-terminus of RAR gamma-1. This peptide (Cat. # PEP-228) is available for use in neutralization and control experiments.

          Target Information

          Retinoic acid, a metabolite of vitamin A, is necessary for normal organogenesis but acts as a teratogen at high levels during embryonic and fetal development. Retinoic acid functions through its interaction with the nuclear protein, retinoic acid receptor (RAR). RAR belongs to the steroid and thyroid hormone superfamily of nuclear receptor proteins which exert their effects by binding to specific DNA response elements, thus regulating gene expression in target cells. RAR exists as three major subtypes: alpha, beta and gamma. Two main isoforms of RAR gamma exist: RAR gamma-1 has been found to be predominantly expressed in skin, while RAR gamma-2 has been found early in embryogenesis and in embryonal carcinoma cells. RAR gamma is also known as RARD. The corresponding gene for the retinoic acid receptor gamma 1 is NR1B3.
