Secreted Phosphoprotein 1 (SPP1) recombinant human protein is supplied as a lyophilized powder. This protein is suitable for use in analysis of protein structure. In general, recombinant proteins can also be used as an immunogen, as a protein standard, or in cell biology research applications.

This recombinant protein is expressed from a DNA sequence encoding the pro form of human SPP1 (NP_001035147.1) (Met 1-Asn 314)fused to a polyhistidine tag at the C-terminus.

Activity: The activity of this protein has not been tested.

Formulation: lyophilized in PBS, pH 7.4, 5% mannitol, 5% trehalose, 0.02% Tween®-80.

Reconstitution: Dissolve the protein in sterile double-distilled water to a concentration of 0.2 mg/mL or lower. It is recommended that the protein be aliquoted and used as soon as possible. Store aliquots under sterile conditions at -20°C. Avoid repeated freeze-thaw cycles.

Background
Secreted phosphoprotein 1, is a member of the osteopontinfamily. It is an extracellular structural protein and thereforean organic component of bone. Secreted phosphoprotein 1 is expressed in bone, although it is also expressed in other tissues. Secreted phosphoprotein 1 acts as acytokine which is involved in enhancing production of interferon-gamma and interleukin-12 and reducing production of interleukin-10. It is essential in the pathway that leads to type I immunity. Secreted phosphoprotein 1 has been implicated as an important factor in bone remodeling. Specifically, research suggests it plays a role in anchoring osteoclasts to the mineral matrix of bones. The fact that Secreted phosphoprotein 1 interacts with multiple cell surface receptors which are ubiquitously expressed makes it an active player in many physiological and pathological processes including wound healing, bone turnover, tumorigenesis, inflammation and ischemia. Therefore, manipulation of plasma Secreted phosphoprotein 1 levels may be useful in the treatment of autoimmune diseases, cancer metastasis, osteoporosis and some forms of stress.

Vitronectin (VTN-N) Recombinant Human Protein, Truncated (Gibco™)

Vitronectin (VTN-N) is a recombinant human protein that provides a defined surface for feeder-free culture of human pluripotent stem cells (PSCs). When used with Essential 8™ Medium (A1517001), vitronectin (VTN-N) has been proven to maintain pluripotency and normal growth characteristics in multiple PSC lines. In addition, vitronectin (VTN-N) has been shown to support PSC growth for >50 passages without any signs of karyotypic abnormalities and to maintain the ability of PSCs to differentiate into all three germ line lineages. As published by Guokai Chen et al. (1) in the laboratory of James Thomson, the VTN-N variant of vitronectin supports human pluripotent stem cell attachment and survival better than wild-type vitronectin when used in conjunction with Essential 8™ Medium.

Collagen I Bovine Protein (Gibco™)

Fibronectin Human Protein, Plasma (Gibco™)

Fibronectin is a large, cell surface and plasma protein that exhibits structural and adhesive properties in cell-associated fibrillar matrices. Fibronectin is one of the primary cell adhesion molecules. Human Plasma Fibronectin is isolated from human plasma by affinity chromtography. Fibronectin has a molecular weight of 440,000 and is made up of two 220,000 dalton polypeptides. Human Fibronectin is filtered (0,2UI) and supplied lyophilized.

Performance and Quality Testing
The purity is determined by SDS-PAGE. Bovine and human fibronectins are assayed for their ability to support cell attachment and spreading, and screened to confirm absence of mycoplasma or bacterial contamination.

FN1 Recombinant Human Protein (His Tag)

Fibronectin 1 (FN1), recombinant human protein is supplied as a lyophilized powder. It is suitable for use in protein studies such as protein structure analysis and protein-protein interactions. In general, recombinant proteins can also be used as an immunogen, as a protein standard, or in cell biology research applications.

This recombinant protein was expressed from a DNA sequence encoding the Fragment 2 (Ser 607 - Pro 1265) of human FN1 (CAD91166) fused to a polyhistidine tag at the C-terminus.

Activity: Measured by its binding ability in a functional ELISA. Immobilized mouse APCS at 10 µg/ml(100 µl/well) can bind biotinylated FN1 with a linear range of 0.625 - 5 µg/ml.

Formulation: Lyophilized in 140 mM NaCl, 2.7 mM KCl, 10 mM Na2HPO4, 1.8 mM KH2PO4, pH 7.4, 5% Mannitol, 5% Trehalose, 0.02% Tween 80.

Fibronectin is one of the primary cell adhesion molecules. Bovine Plasma Fibronectin is purified from plasma by affinity chromatography on gelatin-Sepharose and further purified by ion exchange chromatography to remove glycosaminoglycans. Bovine Fibronectin is filtered (0,2 µm) and supplied lyophilized.
