Store lyophilized/reconstituted at -20°C; once reconstituted make aliquots to avoid repeated freeze-thaw cycles. Please remember to spin the tubes briefly prior to opening them to avoid any losses that might occur from material adhering to the cap or sides of the tube.

DCL3 is a nuclear, protein-coding gene in Arabidopsis thaliana that produces a large multidomain RNase III family endoribonuclease of the Dicer-like class. The DCL3 protein is organized in the canonical Dicer architecture, with an N-terminal DExD/H-box helicase module followed by a helicase C region, a DUF283 segment implicated in macromolecular interactions, a PAZ domain that recognizes the termini of double-stranded RNA substrates, and a C-terminal catalytic core containing tandem RNase III domains plus double-stranded RNA-binding domain(s) required for precise duplex cleavage. Functionally, DCL3 is essential for endogenous RDR2-dependent small interfering RNA biogenesis and is specialized for generating predominantly 24-nucleotide small interfering RNAs from double-stranded RNA precursors produced in the RNA-directed DNA methylation pathway; these 24-nucleotide products guide transcriptional silencing and epigenetic regulation of repetitive and heterochromatic loci through downstream effector complexes, and DCL3 activity is also implicated in methylation-mediated defense against DNA viruses via the same silencing machinery.
