          HYOU1 Antibody (MA5-45339) in ICC/IF

          Immunocytochemistry/Immunofluorescence analysis using human neuroblastoma cells. Fixation involved 4% Formaldehyde for 15 min at RT. Samples were incubated with GRP170 monoclonal antibody (Product # MA5-45339) at 1:100 for 60 min at RT, followed by Goat Anti-Mouse ATTO 488 at 1:100 for 60 min at RT. Counterstain used was Phalloidin Texas Red F-Actin stain; DAPI (blue) nuclear stain at 1:1,000; 1:5,000 for 60 min RT, 5 min RT. Localization: Endoplasmic Reticulum, Endoplasmic Reticulum Lumen.           Product Specific Information

          1 µg/mL of MA5-45339 was sufficient for detection of GRP170 in 20 µg of HEK293 lysate by colorimetric immunoblot analysis using Goat anti-mouse IgG:HRP as the secondary antibody.|Detects approximately 170kDa.

          Target Information

          HYOU1 belongs to the heat shock protein 70 family. The protein is thought to play an important role in protein folding and secretion in the ER. Since suppression of the protein is associated with accelerated apoptosis, it is also suggested to have an important cytoprotective role in hypoxia-induced cellular perturbation. This protein has been shown to be up-regulated in tumors, especially in breast tumors, and thus it is associated with tumor invasiveness. This signal peptide-lacking protein, which is only 3 amino acids shorter than the mature protein in the ER, is thought to have a housekeeping function in the cytosol. In rat, this protein localizes to both the ER by a carboxy-terminal peptide sequence and to mitochondria by an amino-terminal targeting signal.
