SHANK1 Antibody (MA5-45447) in ICC/IF
Immunocytochemistry/Immunofluorescence analysis using human neuroblastoma cells. Fixation involved 4% PFA for 15 min. Samples were incubated with SHANK1 monoclonal antibody (Product # MA5-45447) at 1:50 for overnight at 4°C with slow rocking, followed by AlexaFluor 488 at 1:1,000 for 1 hour at RT. Counterstain used was Phalloidin-iFluor 647 (red) F-Actin stain; Hoechst (blue) nuclear stain at 1:800, 1.6mM for 20 min at RT. (A) Hoechst (blue) nuclear stain. (B) Phalloidin-iFluor 647 (red) F-Actin stain. (C) SHANK1 antibody (D) Composite.

1 µg/mL of MA5-45447 was sufficient for detection of Shank1 in 10 µg of rat brain lysate by colorimetric immunoblot analysis using Goat anti-mouse IgG:HRP as the secondary antibody.|Detects approximately 190-220 kDa (alternative splice variants). No cross-reactivity against Shank2 or Shank3.
Target Information
Seems to be an adapter protein in the postsynaptic density (PSD) of excitatory synapses that interconnects receptors of the postsynaptic membrane including NMDA-type and metabotropic glutamate receptors via complexes with GKAP/PSD-95 and Homer, respectively, and the actin-based cytoskeleton. May play a role in the structural and functional organization of the dendritic spine and synaptic junction.
