Bioreactors are used to culture cells including mammalian (e.g. CHO, CHO-K1), insect, yeast, and bacterial to produce therapeutic proteins, monoclonal antibodies, nanobodies, vaccines, and gene therapies.
Benchtop bioreactors support smaller volumes and include glass, rocker (or wave), and small single-use bioreactors. Glass bioreactors (1 –15 L) are re-usable and are compatible with suspension and adherent cells. Rocker or wave bioreactors typically use single-use bags on a rocker platform; common volumes are 5 – 25 L. For suspension cells, the Thermo Scientific DynaDrive Single-Use Bioreactor 5L is a feature rich benchtop option that easily scales with other DynaDrive Single-Use Bioreactors.
Bioreactors support scale-up by providing a consistent, controlled environment that can be translated from small-scale development to full commercial production. During process development, key parameters—such as pH, dissolved oxygen, temperature and agitation—are optimized at bench scale and then replicated in larger systems using comparable design and control strategies. Modern bioreactors, including the DynaDrive family of single-use bioreactors, are engineered with automation and advanced sensors to maintain similar performance characteristics across volumes.
Mixing, oxygen transfer, turndown ratio, bag quality, controllers, and even tubing connectors can influence bioreactor performance. Mixing and oxygen transfer are important for cell health and consistent results. A high turndown ratio reduces the number of seed train steps improving yields and productivity. Compatible bags must be of films that are safe for cells with low leachables and extractables. It is also important that the bag manufacturer has a robust and redundant supply chain. Look for details like the Thermo Scientific BioTitan Rentention Device which prevents leaks at tubing connection points.
Single-use bioreactors significantly reduce contamination risk since there is no direct contact between the bioprocessing fluids and the bioreactor. They also reduce cleaning steps, leading to faster equipment turnaround time and great lab productivity. Single-use bioreactors are typically smaller and modular, reducing capital equipment expenditures and the need for facility build outs.
