Thermo Scientific Lambda DNA/HindIII Marker is recommended for sizing of linear double-stranded large DNA fragments in agarose gels. Lambda DNA is digested to completion with the appropriate Thermo Scientific restriction enzyme(s) and purified and dissolved in storage buffer. DNA fragments contain various sticky ends depending on the restriction enzyme used for preparation of the marker.

The marker can be labeled radioactively with T4 Polynucleotide Kinase. Alternatively, and it can be labeled radioactively or non-radioactively with Klenow Fragment or Klenow Fragment, exo- with the fill-in reaction (see Resources for Protocols).

Note
The cohesive ends (the 12 nt cos site of lambda DNA) of fragments indicated may anneal and form additional bands. These fragments can be separated by heating at 65°C for 5 minutes and then cooling on ice for 3 minutes.
