Thermo Scientific Pierce Sulfo-SMCC formato No-Weigh es un entrecruzador de amina a sulfhidrilo soluble en agua que contiene grupos reactivos de éster NHS y maleimida en extremos opuestos de un brazo separador de ciclohexano de longitud media (8,3 angstroms).

Características de Sulfo-SMCC, formato No-Weigh:

Grupos reactivos: Éster Sulfo-NHS y maleimida
Reactivo a: grupos amino y sulfhidrilo
• Soluble en agua (en comparación con SMCC), por lo que el entrecruzamiento se puede hacer en soluciones fisiológicas

• El puente de ciclohexano confiere mayor estabilidad al grupo de maleimida haciendo de Sulfo-SMCC un agente de entrecruzamiento ideal para la activación de maleimida de proteínas. Los grupos de maleimida son estables durante 64 horas en tampón fosfato de sodio 0,1 M, pH 7 a 4 °C.

Sulfosucinimidil-4-(N-maleimidometilo)ciclohexano-1-carboxilato (Sulfo-SMCC) es un entrecruzador no escindible e impermeable a la membrana. Contiene una N-hidroxisucinimida reactiva a la amina (éster NHS) y un grupo de maleimida reactiva a sulfhidrilo. Los ésteres NHS reaccionan con aminas primarias a pH 7-9 para formar enlaces de amida estables. Las maleimidas reaccionan con grupos sulfhidrilo a pH 6,5-7,5 para formar enlaces tioetéter estables. Los grupos de maleimida de Sulfo-SMCC y SMCC son inusualmente estables hasta un pH 7,5 debido al puente ciclohexano en el brazo separador. Dado que contiene la fracción sulfonilo hidrófila, Sulfo-SMCC es soluble hasta en ∼10 mm de agua y muchos de los tampones más utilizados. What is the difference between SMCC and Sulfo-SMCC?

SMCC is an amine-to-sulfhydryl crosslinker that contains NHS-ester and maleimide reactive groups at opposite ends of a medium-length cyclohexane-stabilized spacer arm (8.3 angstroms). Sulfo-SMCC is a similar compound but it has a sulfonate group on the NHS ring which makes it water-soluble whereas SMCC is water-insoluble and needs to be dissolved first in an organic solvent like DMF or DMSO. Subsequent dilution into aqueous reaction buffer is generally possible, and most protein reactants will remain soluble if the final concentration of organic solvent is less than 10%.
