Alkaline Phosphatase staining of iPSC Colonies

(A-B) 200,000 primary human fibroblasts were reprogrammed with (A) the CytoTune-iPS Sendai Reprogramming Kit and (B) lentivirus. Twenty-eight days post reprogramming, ~245 colonies were observed from CytoTune reprogramming, while the lentivirus reprogramming generated ~10 colonies. (C-D) 50,000 primary human fibroblasts were reprogrammed with (C) the CytoTune-iPS Sendai Reprogramming Kit and (D) mRNA. Twenty-eight days post-reprogramming, ~130 iPSC colonies were observed from CytoTune reprogramming, while 11 iPSC colonies were observed from mRNA reprogramming.
