The NanoDrop Eight and NanoDrop Ultra Spectrophotometer and Fluorometers come with Acclaro Sample Intelligence Technology out of the box. This software feature will notify the user of any contamination or irregularities in most of NanoDrop instruments’ native applications. Thermo Fisher Scientific does offer an optional software package called Acclaro Pro Software that includes new versions of the most popular measurement applications. This new software is completely different than the Acclaro Sample Intelligence Technology and offers as little as ±5% error throughout the dynamic absorbance range.
Acclaro Sample Intelligence Technology is ideal for day to day usage. It can help with contamination identification, identify bubbles, and notify the user when absorbance ratios are outside acceptable ranges. When the software identifies a contaminant, it notifies users by showing a yellow contaminant icon next to the sample. Selecting the yellow icon displays the original sample, the contaminant, and the corrected analyte spectra. A table reports the corrected concentration, so the user knows what contaminant is present and whether to use the sample in downstream experiments. The rest of the analysis can then be continued with confidence. The Acclaro Sample Intelligence Technology feature is available in the following applications: dsDNA, RNA, and Protein A280.
The Acclaro Pro Software is suited for advanced applications in biopharma such as production and manufacturing with high-precision applications; protein and antibody manufacturing; vaccine production; and biotechnology for protein and oligonucleotide manufacturing. Acclaro Pro software offers even more comprehensive features and capabilities within the expanded application suite. Acclaro Pro provides seven standalone software applications: dsDNA pro, ssDNA pro, RNA pro, Protein A280 pro, Oligo DNA pro, Oligo RNA pro, and Custom Factor pro.
Acclaro Contaminant Identification uses sophisticated mathematical algorithms to identify contaminants in samples such as phenol, guanidine salts, and protein in nucleic acids. Based on the full absorbance spectrum, it quantifies the contaminant’s contribution to the total sample absorbance and calculates the true analyte concentration.

Purity ratios alone cannot give an accurate picture of sample contamination. Some common contaminants like phenol absorb at 260 nm, the same as nucleic acids. As shown in the video, phenol contamination can cause other UV-Vis instruments to overestimate nucleic acid quantity, resulting in elevated PCR Ct values due to inadequate template. Acclaro Sample Intelligence Technology recognizes this, and allows NanoDrop instruments to identify and correct for a broad range of contaminants.

RNA contamination in genomic DNA preparations is a common issue in molecular biology workflows. Traditionally, UV absorbance technology cannot distinguish between DNA and RNA, since both absorb light at 260 nm. When measuring samples with traditional spectrometry, copurified RNA will artificially inflate the concentration of DNA, causing potential errors in downstream applications.

Back in the “bad old days” BND—Before NanoDrop Spectrophotometers—quantitating nucleic acid samples was a slow and laborious process. Highly concentrated samples had to be diluted and pipetted into cuvettes for measurement. The protocols required time, expert knowledge, tedious dilutions, and complicated workflows—and were prone to errors and replicability issues.
In 2001, the NanoDrop 1000 Spectrophotometer revolutionized UV-Vis analysis, allowing for accurate quantitation of only 1–2 µL of sample without cuvettes or capillaries.
