The Applied Biosystems QuantStudio 6 and 7 Pro real-time PCR systems deliver greater flexibility to support a variety of applications and throughput requirements. The systems’ smart features are designed to reduce contamination by allowing you to set up your run hands-free. Schedule and monitor your run from your mobile device, set-up your experiment, analyze data, and share your results from your computer no matter where it is or where you are.  
  

The QuantStudio 6 and 7 Pro systems’ 96-well blocks incorporate VeriFlex technology for precise temperature control. These blocks are divided into isolated zones that allow separate cycling conditions within the same run -- perfect for optimization. You can also increase your throughput with interchangeable blocks that accommodate 96-well 0.2 ml blocks, 96-well 0.1 ml blocks, 384-well blocks and TaqMan Array Cards (QuantStudio 7 Pro model only). Other unique features of the QuantStudio 7 Pro system include automation compatibility and software features to assist with 21 CFR Part 11 compliance. Distinct clusters indicating 2-fold and 1.5-fold discrimination on QuantStudio 7 Pro system with 99.9% confidence, equivalent to other Applied Biosystems platforms, QuantStudio 7 Flex and the 7900HT instruments. Total liver cDNA samples with 2-fold and 1.5-fold differences in input were assessed. Runs were analyzed using their respective platform software, with auto-threshold and baseline, then exported and analyzed using SAS JMP v13 statistical software.

The QuantStudio 7 Pro system can detect 1.5 fold changes in expression. A dilution series of the KAZ plasmid was amplified. The vertical axis is the change in fluorescence normalized to the ROX dye. The horizontal axis is increasing cycle number from left to right. This is verified by plotting as a standard curve on the right. The vertical axis of the standard curve is the cycle threshold value, and the horizontal axis is the dilution series increasing in concentration from left to right.

Comparable results across QuantStudio 7 Pro system and other Applied Biosystems platforms when comparing different genes. To demonstrate equivalency of relative gene expression results using the new QuantStudio 7 Pro system compared to those using the QuantStudio 7 Flex and 7900HT instruments, three gene assays (GFAP, ALB, and KRT1) labeled with FAM-MGB and an endogenous control, PPIA (VIC-QSY, primer limited), were run on each platform. 

Highly reproducible separation over the 10 logs of input template amount demonstrated by amplification of KAZ plasmid DNA in 10-fold dilutions using the 96-well block was performed. The vertical axis is the log of the background subtracted fluorescence normalized to the ROX dye. The horizontal axis is increasing cycle number from left to right.

Single copy of the KAZ plasmid in blue compared to NTC in purple.  The QuantStudio 7 Pro system consistently detects a single copy, clearly distinguishable from NTCs (43/48 positive wells).

Multiplex reaction detects 18S rRNA labeled with VIC dye and TGFb  gene with FAM labeled TaqMan probes and primers. The reaction used the fast master mix and was run in 96 replicates. The vertical axis reflects the change in florescence normalized to the ROX dye, and the horizontal axis reflects increasing cycles from left to right. The CT values show a clear, consistent separation of products with a low standard deviation among replicates.
