How to use the EZPassage tool for cell passaging?
- Under a laminar-flow hood, inspect the culture and remove any differentiated areas if present.
- Remove the packaging of the EZPassage tool and hold the culture dish in one hand.
- First rolling pass: With the tool in contact with the plate surface (still containing medium), roll the tool across the dish in one direction (left to right), applying enough pressure to engage the roller blades.
- Second rolling pass: Rotate the dish 90° and roll again in the new direction. This produces a grid-pattern cut across the colonies, generating uniformly sized colony fragments.
- Using the serological pipette, add fresh medium to the dish and gently flush the plate surface to dislodge the cut fragments into suspension.
- Transfer the colony fragments to a conical tube; spin briefly (e.g., 1,000 rpm ≈ 200 × g for 2 minutes) or allow gravity-settling.
- Aspirate supernatant carefully to remove single cells or unwanted feeder cells if present and resuspend the colony fragments in fresh medium.
- Seed the colony fragments onto the prepared new plate at the desired split ratio (e.g., 1:2 to 1:4) and place in incubator. Avoid moving the plate for at least 24 h to allow attachment.
- Discard the EZPassage tool—do not reuse.
