Thermo Scientific Tomography 5 Software supports tomographic data acquisition across diverse life sciences and materials science samples, including resin-embedded and cryogenically prepared samples. From investigating cellular structures and macromolecular complexes to characterizing nanoscale structures in 3D with full chemical and structural context, it offers the versatility and ease of use needed for advanced research.

 

Tomography 5 Software supports flexible cryo-electron tomography workflows across a range of transmission electron microscopes, including the Thermo Scientific™ Talos™ 12 TEM, Thermo Scientific™ Spectra™ TEM, and other systems without an Autoloader. As part of the Thermo Scientific™ CryoTomo Software Suite, it adds advanced automation and tilt-series acquisition and analysis capabilities to enable higher-throughput cryo-electron tomography on Thermo Scientific™ Glacios™ and Krios™ Cryo-TEMs.

Features of Tomography 5 Software for TEM 3D data acquisition

The Atlas is a low-magnification map of each grid, which makes it easy to assess grid quality, screen grids, and select sites for high-magnification imaging.

Search Maps are sets of tiles with medium magnification that allow you to map entire grid squares. They help increase data collection throughput and improve navigation accuracy.

Multisite batch tomography streamlines automated data acquisition and boosts TEM throughput. Simply identify your regions of interest, configure the tilt series, and let the system take over. Once parameters are set, acquisition proceeds fully autonomously

Off-axis multi-shot acquisition significantly enhances throughput. It records several tomogram positions with only one focus and tracking area, streamlining data acquisition, and increasing efficiency.

The dose symmetric acquisition scheme optimally distributes the electron dose for high-resolution sub-tomogram averaging. During a tomographic tilt series, the acquisition scheme exposes the projections in the lower negative and positive angular tilt range uniformly, preserving high-resolution information. This collection scheme optimizes resolution (i.e., the sub-nanometer range) and provides considerably better results than continuous or bi-directional tilt schemes.

Multiple cameras and detectors enable a wide range of use cases and increase versatility. Tomography 5 Software is fully integrated with Thermo Scientific Ceta CMOS Cameras, Thermo Scientific Falcon 4i Direct Electron Detectors, Panther STEM Detectors, and Ultra-X EDX Detectors.

The Thermo Scientific Selectris X Imaging Filter greatly improves the signal-to-noise ratio  in tomography, boosting contrast and resolution through zero-loss filtering (i.e. the removal of inelastically scattered electrons). The imaging filter is fully integrated with Tomography 5 Software.
