Do you have a favorite list of genes you would like to target with a collection of siRNAs? We can prepare a custom Silencer Select siRNA library for any set of human genes. The minimum order is only 36 siRNAs. All you need to supply is a list of genes or transcripts identified by NCBI Entrez Gene ID or RefSeq mRNA accession number. With this user-friendly option, you can specify the order of the siRNAs on the plate, as well as the amount of siRNA per order (1 nmol minimum), number of aliquots on each plate, and the number of siRNAs per target. siRNAs can be provided individually or pooled.
Individual siRNAs for enhanced data reliability
Silencer Select siRNA Libraries feature three individual siRNAs for each target. Screening with three siRNAs per gene significantly decreases both false positive and false negative rates, resulting in enhanced confidence in RNAi screening data, reduced chance of missing important genes, and less time spent following up on false positive hits from the screen.
These libraries contain 0.25 nmol of each siRNA, sufficient for 500 transfections (at 5 nM siRNA, 100 µL transfection volume). The siRNAs are supplied dried into the wells of 96 well plates. One column of each 96 well plate is left empty to allow for the addition of controls at the time of use. The libraries are delivered with full siRNA sequence information, as well as the exon targeted and each targeted gene's symbol, full name, aliases, RefSeq number, and Entrez Gene ID, plus results from any validation experiments performed using the included siRNAs.
Silencer Select siRNA Libraries to these or any other list of human genes can also be provided with 1, 2, or 5 nmol of each siRNA. Custom formatting is available with these larger sizes. We guarantee that when you purchase two Silencer Select Pre-designed siRNAs to the same target, then those two siRNAs will silence the target mRNA by 70% or more. To qualify for the guarantee, the siRNAs must have been transfected at ≥5 nM and mRNA levels detected 48 hours post-transfection. Real-time RT-PCR is recommended but not required for this application. Customers must also show sufficient knockdown with a positive control siRNA to demonstrate transfection efficiency. If the guaranteed level of knockdown is not observed and an appropriate positive control is successful, a new Silencer Select siRNA sequence will be synthesized free of charge.
