Fast, reproducible labeled n-glycan analysis

Released glycans have no chromophore and a poor response with conventional UV detection. They can be labeled with fluorescent and/or mass spectrometric tags prior to high sensitivity analysis by fluorescence detection, typically using 2-aminobenzamide (2-AB).

Thermo Scientific aminoxyTMT™ (Tandem Mass Tag™) reagents can also be used for labeling released glycans. This enables multiplexing experiments to be performed for quantitation of glycans by mass spectrometry.

  • Thermo Scientific GlycanPac™ AXR1 LC columns. This high-performance, silica based chromatography column achieves mixed mode separation using a combination of charge, isomer structure, and size. Now there's a column with unique selectivity to analyze biologically important glycans.
  • For glycans that contain ionizable groups, a mixed mode HILIC/anion exchange column like the Thermo Scientific GlycanPac™ AXH-1 column demonstrates a complementary solution for the analysis of charged glycans, with separation based on charge in addition to hydrophilicity.
  • Hydrophilic interaction liquid chromatography (HILIC) features increased retention of polar species and has been shown to give good retention of  oligosaccharides. The Thermo Scientific Accucore 150-Amide-HILIC HPLC phase is designed for the separation of hydrophilic biomolecules.
