The Invitrogen Virus Precipitation Reagent and Dynabeads Intact Virus Enrichment kit allows for multiple options to get the enriched virus sample you need. The Invitrogen Intact Virus Precipitation Reagent can enrich intact virus from large volume and dilute samples by precipitation in three simple steps. You only need a centrifuge, and the intact virus is enriched in less than 3 hours with very little hands-on time.

On the other hand, if you are looking for a fast and efficient automation solution, KingFisher instruments can be combined with Dynabeads Intact Virus Enrichment beads for processing of up to 96 virus-containing samples in only 20 minutes. If preferred, the intact virus can easily be removed from the beads in a short 10 min release step. The Dynabeads can efficiently isolate SARS-CoV-2 from cell culture media, virus transport media and wastewater samples.

Product principle Reagent that precipitates the virus from solution by tying up water molecules, which results in less-soluble components (i.e., virus) being forced out of solution. The virus is collected after a brief, low-speed centrifugation. 1 µm-sized magnetic beads with strong anion exchange surface; binds negatively-charged vesicles and molecules.

Both products are optimized for the enrichment of live or inactivated SARS-CoV-2 and SARS-CoV-2 virus–like particles (VLP’s) but these products are generic and can be used for other enveloped viruses or vesicles as well. The enriched virus is fully intact and can be used for functional assays, or downstream end point analysis of viral nucleic acid or protein.

This reagent is designed to precipitate the virus after a short incubation and centrifugation step.

These strong anionic-exchange magnetic Dynabeads allow for fast binding of negatively-charged vesicles and molecules to the beads. This facilitates automation using any of our KingFisher instruments for either a few samples or up to 96 samples/run, depending on the instrument.

Intact Virus Precipitation Reagent (in 3 simple steps)

Use 2X volume of virus sample to precipitation reagent (e.g., use 2 mL virus sample for every 1 mL reagent).

  •  Add precipitation reagent to the virus sample, and incubate for 2 hours at 2–8 °C
  •  Centrifuge (30 min) at 10,000 x g
  •  Remove supernatant and keep the pellet with the enriched virus

The enriched intact virus is now ready for downstream analysis and can be used directly in a PCR reaction without any further nucleic acid extraction.


Dynabeads Intact Virus Enrichment (<96 samples in 20 minutes)

Use ~20 µL beads per 1 mL virus sample. The protocol is for 20 µL beads per 1 mL sample, but the automated script has half of the reagent volumes (10 µL beads per 200-500 µL virus sample). The protocol can be scaled up or down by scaling all volumes accordingly. The washing and release buffers are described in the user guide.

Automated

The automated protocol is based on the KingFisher instruments, software, and plastic. KingFisher Duo Prime holds 12 samples; KingFisher Flex holds 24-well plates and 96-well plates.

  •  Add the buffers, Dynabeads, and virus samples to the plates
  •  Push the Start button and come back in 20 min to collect your enriched virus!
  •  Optional: Release the virus from the beads in 10 min

The enriched virus is now ready for downstream analysis.

For downstream qRT-PCR we recommend using:

  • MagMAX™ Viral/Pathogen Nucleic Acid Isolation Kit when the virus is enriched from cell culture media or virus transport media
  • MagMAX™ Microbiome Ultra Nucleic Acid Isolation Kit when the virus is enriched from wastewater samples

 Manual

A mixer/roller and a DynaMag magnet is required for the washing steps. Mix the beads thoroughly before you start.

  •  Wash the beads once
  •  Add beads to sample for 10 min at RT
  •  Wash the bead-bound virus once with a magnet to remove unbound target
  •  Optional: Release the virus from the beads in ~10 min

The enriched virus is now ready for downstream analysis.
