Ion AmpliSeq™ technology delivers simple and fast library construction for affordable targeted sequencing of specific human genes or genomic regions. Based on ultrahigh-multiplex PCR, Ion AmpliSeq™ technology requires as little as 10 ng of input DNA to target sets of genes, making sequencing of FFPE samples routine on Ion PGM™ Systems.

Alternative target selection methods are lengthy and complex and require large amounts of DNA, making them impractical for sequencing DNA from the formalin-fixed, paraffin-embedded (FFPE) tissues that are commonly used in translational and clinical research. Using just 10 ng of FFPE DNA and one pool of 6,144 primer pairs, variants can be identified in a single day using the Ion AmpliSeq™ Custom workflow. By contrast, the same project using alternative workflows requires 100 times as much DNA, 8 times the pools, and twice the time.

Ion TargetSeq™ Exome Kits offer a simple and flexible workflow that provides single tube enrichment using solution-phase high-density DNA probe capture technology for highly specific enrichment of exons and other targeted regions within the human genome. Ion TargetSeq™ Exome Kits utilize the highest capture probe density (> 2M biotinylated capture probes) along with a probe tiling strategy that provides exceptional capture efficiency resulting in high “on target” sequencing read percentages. The kits combine the advantages of low DNA input, as little as 125 ng of each individual library, with speed - 2.5 day library preparation and capture – that facilitate sample multiplexing with up four barcoded samples in a single target enrichment reaction. The capture protocol and capture probe content have been thoroughly optimized to give superior performance on both Ion Torrent PGM and Proton sequencing instruments. Further, the exome sequencing workflow leverages the enzymatic sheering protocol of Ion Express™ Plus Fragment Library kit for integrated workflow that permits easy automation.
